    1. cancer gdc-0449

GDC-0449 is a drug used to treat advanced basal cell carcinoma that has spread to other parts of the body or has come back after surgery. It is also used in patients who cannot be treated with surgery or radiation therapy. It is also being studied in the treatment of other types of cancer. GDC-0449 blocks a type of protein involved in tissue growth and repair and may block the growth of cancer cells. It is a type of Hedgehog signaling pathway antagonist. Also called Erivedge and vismodegib. Cancer is a term for diseases in which abnormal cells divide without control and can invade nearby tissues. Cancer cells can also spread to other parts of the body through the blood and lymph systems. There are several main types of cancer. Carcinoma is a cancer that begins in the skin or in tissues that line or cover internal organs. Sarcoma is a cancer that begins in bone, cartilage, fat, muscle, blood vessels, or other connective or supportive tissue. Leukemia is a cancer that starts in blood-forming tissue such as the bone marrow, and causes large numbers of abnormal blood cells to be produced and enter the blood. Lymphoma and multiple myeloma are cancers that begin in the cells of the immune system. Central nervous system cancers are cancers that begin in the tissues of the brain and spinal cord. Also called malignancy.

      • For example, PURPOSE This phase I trial is studying the side effects and best dose of erlotinib hydrochloride when given together with GDC (General Dental Council (UK))-0449 with or without gemcitabine hydrochloride in treating patients with metastatic pancreatic cancer or solid tumors that cannot be removed by surgery. Pancreatic cancer is a disease in which malignant (cancer) cells are found in the tissues of the pancreas. Also called exocrine cancer. Metastatic, having to do with metastasis, which is the spread of cancer from the primary site (place where it started) to other places in the body. Erlotinib hydrochloride is a drug used to treat certain types of non-small cell lung cancer. It is also used together with gemcitabine to treat pancreatic cancer and is being studied in the treatment of other types of cancer. Erlotinib hydrochloride is a type of epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. Also called CP-358,774, erlotinib, OSI-774, and Tarceva. Surgery is a procedure to remove or repair a part of the body or to find out whether disease is present. An operation. Phase I trial is the first step in testing a new treatment in humans. These studies test the best way to give a new treatment (for example, by mouth, intravenous infusion, or injection) and the best dose. The dose is usually increased a little at a time in order to find the highest dose that does not cause harmful side effects. Because little is known about the possible risks and benefits of the treatments being tested, phase I trials usually include only a small number of patients who have not been helped by other treatments. Gemcitabine is the active ingredient in a drug that is used to treat pancreatic cancer that is advanced or has spread. It is also used with other drugs to treat breast cancer that has spread, advanced ovarian cancer, and non-small cell lung cancer that is advanced or has spread. It is also being studied in the treatment of other types of cancer. Gemcitabine blocks the cell from making DNA and may kill cancer cells. It is a type of antimetabolite Gemcitabine hydrochloride is a drug used to treat pancreatic cancer that is advanced or has spread. It is also used with other drugs to treat breast cancer that has spread, advanced ovarian cancer, and non-small cell lung cancer that is advanced or has spread. It is also being studied in the treatment of other types of cancer. Gemcitabine hydrochloride blocks the cell from making DNA and may kill cancer cells. Also called Gemzar. Erlotinib is a drug used to treat certain types of non-small cell lung cancer. It is also used together with gemcitabine to treat pancreatic cancer and is being studied in the treatment of other types of cancer. Erlotinib is a type of epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. Also called CP-358,774, erlotinib hydrochloride, OSI-774, and Tarceva. Pancreatic, having to do with the pancreas.

      • Evidently, to assess the effect of erlotinib hydrochloride and Hedgehog antagonist GDC-0449 on fludeoxyglucose F 18 positron emission tomography imaging in patients with metastatic pancreatic cancer. Tomography is a series of detailed pictures of areas inside the body. The pictures are created by a computer linked to an x-ray machine. Fludeoxyglucose F 18 is the radioactive form of glucose used in positron emission tomography (PET), a diagnostic imaging procedure. Antagonist is in medicine, a substance that stops the action or effect of another substance. For example, a drug that blocks the stimulating effect of estrogen on a tumor cell is called an estrogen receptor antagonist. Imaging is in medicine, a process that makes pictures of areas inside the body. Imaging uses methods such as x-rays (high-energy radiation), ultrasound (high-energy sound waves), and radio waves.

      • One can view, to assess the effect of erlotinib hydrochloride and Hedgehog antagonist GDC-0449 on selected biomarkers in circulating tumor cells and tumor biopsy samples from patients with metastatic pancreatic cancer. Biopsy is the removal of cells or tissues for examination by a pathologist. The pathologist may study the tissue under a microscope or perform other tests on the cells or tissue. There are many different types of biopsy procedures. The most common types include: (1) incisional biopsy, in which only a sample of tissue is removed; (2) excisional biopsy, in which an entire lump or suspicious area is removed; and (3) needle biopsy, in which a sample of tissue or fluid is removed with a needle. When a wide needle is used, the procedure is called a core biopsy. When a thin needle is used, the procedure is called a fine-needle aspiration biopsy.
